Ze worden vaak genoemd in discussies over veroudering omdat GH- en IGF-1-patronen met leeftijd veranderen. Dat betekent echter niet dat het verhogen van GH\/IGF-1 automatisch gezond ouder worden of een langere levensduur veroorzaakt.<\/p>\n\n\n\n<blockquote class=\"wp-block-quote is-layout-flow wp-block-quote-is-layout-flow\"><p><strong>Bewijsstatus:<\/strong> Humane studies met secretagogen laten endocriene en soms lichaamssamenstellings-effecten zien, maar er is geen overtuigend bewijs dat deze middelen de menselijke levensduur verlengen. De balans tussen GH\/IGF-1, metabolisme, functie en ziekte is complex.<\/p><\/blockquote>\n\n\n\n<h2 class=\"wp-block-heading\">Twee belangrijke farmacologische routes<\/h2>\n\n\n\n<h3 class=\"wp-block-heading\">GHRH-analogen<\/h3>\n\n\n\n<p class=\"wp-block-paragraph\">GHRH-analogen stimuleren de hypofyse via de groeihormoon-releasing-hormoonreceptor. CJC-1295 en tesamorelin behoren tot deze onderzoekscategorie. De gepubliceerde humane CJC-1295-studies onderzochten een <strong>langwerkende<\/strong> vorm. Die resultaten mogen niet automatisch worden toegeschreven aan zogenoemd CJC-1295 \u201czonder DAC\u201d.<\/p>\n\n\n\n<h3 class=\"wp-block-heading\">Ghreline\/GHSR-agonisten<\/h3>\n\n\n\n<p class=\"wp-block-paragraph\">Middelen zoals MK-677 en capromorelin activeren de ghrelinreceptor\/GHSR-route. In gerandomiseerde studies bij oudere volwassenen stegen GH\/IGF-1 en veranderden sommige lichaamssamenstellings- of functionele maten. Dat leverde geen bewijs voor levensduurverlenging en sommige studies rapporteerden metabole nadelen, waaronder ongunstige veranderingen in glucosehuishouding.<\/p>\n\n\n\n<h2 class=\"wp-block-heading\">CJC-1295<\/h2>\n\n\n\n<p class=\"wp-block-paragraph\">In gezonde volwassenen verhoogde langwerkend CJC-1295 GH en IGF-1 gedurende meerdere dagen. Dit is farmacodynamisch bewijs, geen longevityonderzoek. De studie ondersteunt evenmin claims over CJC zonder DAC, combinaties met ipamorelin of langdurig gebruik bij gezonde mensen.<\/p>\n\n\n\n<h2 class=\"wp-block-heading\">Ipamorelin<\/h2>\n\n\n\n<p class=\"wp-block-paragraph\">Ipamorelin is in vroege humane farmacokinetische\/farmacodynamische studies onderzocht en later in een postoperatieve indicatie. Die onderzoeken tonen geen effect op menselijke healthspan of levensduur.<\/p>\n\n\n\n<h2 class=\"wp-block-heading\">Tesamorelin<\/h2>\n\n\n\n<p class=\"wp-block-paragraph\">Tesamorelin heeft een offici\u00eble geneesmiddelenstatus, maar voor een specifieke klinische toepassing: reductie van excessief abdominaal vet bij volwassenen met HIV-geassocieerde lipodystrofie. De indicatie is geen goedkeuring voor anti-aging of longevity.<\/p>\n\n\n\n<h2 class=\"wp-block-heading\">Waarom \u201cstack\u201d- of combinatieclaims niet thuishoren in een wetenschappelijk kenniscentrum<\/h2>\n\n\n\n<p class=\"wp-block-paragraph\">Combinaties zoals CJC-1295 plus ipamorelin worden online veel genoemd, maar populariteit is geen klinisch bewijs. Zonder gerandomiseerde combinatiestudies kunnen werkzaamheid en veiligheid van zo\u2019n combinatie niet worden afgeleid uit afzonderlijke studies.<\/p>\n\n\n\n<h2 class=\"wp-block-heading\">Conclusie<\/h2>\n\n\n\n<p class=\"wp-block-paragraph\">Secretagogen zijn relevante onderzoeksmoleculen voor endocrinologie en lichaamssamenstelling, maar de huidige humane literatuur ondersteunt geen algemene longevityclaim.
